Sensitivity-improved immunoassay for SARS-CoV-2 spike protein in saliva without pretreatment by using immuno-wall microfluidic device

Abstract

In previous publication, we reported a sandwich-shape immunoassay for SARS-CoV-2 spike protein in saliva without pretreatment by using an immuno-wall microfluidic device (IMD). The assay time was 30 min, and the limit of detection (LOD) was 10 - 50 ng/mL. In this study, we developed a method to improve the sensitivity of IMD by using a designed automatic microfluidics delivery system for analyte enrichment. Through this method, the assay time was reduced to about 20 min and the LOD was improved to less than 1 ng/mL. The improved IMD can be applied to the rapid and ultrasensitive test for COVID-19 in the future.
