The combined effects of polaprezinc and sucralfate on various experimental models of gastric lesions, were investigated in rats. Polaprezinc and sucralfate dose dependently prevented the development of gastric lesions induced by ammonia or indomethacin. The ED50 of polaprezinc and that of sucralfate for suppressing gastric lesions were remarkably reduced when these drugs were used in combination. ED50 of polaprezinc and sucralfate in ammonia induced lesions were equally the theoretical values calculated on the basis of additive action. The synergistic effect of polaprezinc and sucralfate in suppressing of indomethacin induced lesions were observed. Polaprezinc significantly increased zinc contents of ulcer region in acetic acid ulcer. The enhancement of zinc contents by polaprezinc was not influenced by the combine treatment of sucralfate. These marked synergistic potentiation of polaprezinc and sucralfate may be due to compensatory coordination of both drugs on the difference of actions. These results suggest that the combination of polaprezinc and sucralfate may be beneficial for the treatment of gastric lesions in clinical therapy.
