We investigated the anti-ulcer effects of polaprezinc (Z-103) and sucralfate using several acute experimental models of gastric and duodenal lesions in rats. Z-103 prevented the development of gastric and duodenal lesions induced by water immersion stress, acidified ethanol, indomethacin, compound 48/80 and mepirizole. However, Z-103 was not influenced on experimental models of gastric lesions related to acid secretion, such as aspirin in pylorus-ligated and Shay ulcers. Sucralfate also prevented the development of gastric and duodenal lesions on several experimental models except for compound 48/80. The efficacy of Z-103 was superior to sucralfate, without aspirin in pylorus-ligated and Shay ulcers. These results indicate that Z-103 strongly prevents the development of gastric and duodenal lesions without affecting acid secretion. Therefore, Z-103 may be useful for the treatment of gastric and duodenal ulcers in clinical therapy.
