We attempted to investigate interaction of phencyclidine (PCP) on serotonin1 (5-HT1) and serotonin2 (5-HT2) receptors in rat brain synaptic membrane since some of PCP-induced stereotyped behaviors were similar to 5-HT dependent behaviors in rats. PCP inhibited strongly the binding of 5-HT2 receptor antagonist, [3H]-spiperone in the presence of dopamine receptor antagonist, sulpiride, at the IC(50) of 14.8 nM. However, in terms of 5-HT1 agonists, 5-methoxy-N,N-dimethyl-tryptamine and 5-HT the dose range of mM was need to inhibit it. On the contrary, 5-HT inhibited strongly the binding of 5-HT1 receptor agonist, [3H]-5-HT, but PCP did not. These results suggest that PCP interacts with 5-HT2 receptor, but not 5-HT1 receptor.
